Definition of Fradditive™: (1) fragments of a ligand or macromolecule found bound to proteins in nature; (2) very small chemical fragments of known drugs or natural products. 

 

Why Fradditives™?

 

  • Ligands stabilize proteins in solution improving stability and crystallizability

  • Serendipitous contaminants, buffer or crystallization components have been critical for crystallizability 

  • Ligands have not been routinely incorporated into commercial screens

  • 20 years of fragment-based lead discovery data shows that fragments find binding sites on proteins

  • Not all fragments alter protein activity but are relevant to protein stability

Fradditive™ Product Line

 

  • Low molecular weight (< 150) and soluble to 10mM in aqueous solution to improve their probability of binding to your target

  • 96 conditions pre-formulated and ready for use

Fradditive™ Screen 1

  • Fradditive™ Screen 1 is composed of natural and unnatural amino acids, sugars, nucleotide and nucleotide analogues.

  • Amino acids and sugars are well-known to prevent aggregation in solution through non-specific binding.

  • These compounds may bind specifically or non-specifically to your target to stabilize it in solution during purification or crystallization.

  • We recommend screening at a 1:10 dilution.

  • 0.5ml of 96 aqueous solutions are provided in deep well blocks to facilitate high-through sampling

Fradditive™ Screen 2

  • Fradditive™ Screen 2 is composed of very small fragments (<150 MW) of natural products

  • As with Fradditive™ Screen 1, these compounds are expected to bind specifically or non-specifically to your target to stabilize it in solution or prevent aggregation

  • We recommend screening at a 1:10 dilution

  • 0.5ml of 96 aqueous solutions are provided in deep well blocks to facilitate high-through sampling

 

Fradditive™ Screen - pH

  • Sparse matrix screen composed of 32 unique fragments and 32 unique buffer-pH combinations

  • Screen may be used to optimize protein purfication and crystallization

  • pH range that most commonly yield crystals is more highly sampled

  • Fradditive™ are a diverse sampling of the Fradditive™ collection (screens 1 and 2) including amino acids, nucleotides and fragments of natural products

  • We recommend screening at a 1:10 dilution

  • 0.5ml of 96 aqueous solutions are provided in deep well blocks to facilitate high-through sampling

 

Fradditive™ Screen - Salt

  • Sparse matrix screen composed of 32 unique fragments and 24 unique salt combinations

  • Salts can be used to solubilize proteins and at higher concentrations used to yield crystals

  • Screen may be used to optimize protein purfication and crystallization

  • Salts that most commonly yield crystals were chosen for the screen

  • Fradditive™ are a diverse sampling of the Fradditive™ collection (screens 1 and 2) including amino acids, nucleotides and fragments of natural products

  • We recommend screening at a 1:10 dilution
